It depends on your target viewers. If it's the general public then I'm afraid you should design for 800*600 or use max/min width hacks to go down to 800 which is my preference. Lots of people still have old W98 computers on 800*600, including firms who just haven't upgraded because of the cost.
If you know most of your viewers are reasonably up to date with computers then definitely go for 1000px minimum, maybe 1280px if you are really sure they will have that capability.
There is a half-way house often used by businesses - like my isp for example.
They design the most important bit 780px wide fixed left, then have less important stuff like contact links, ads, privacy statements and the like going further out to say 1000 for 1024px resolutions. This means people with 800*600 still see the important stuff without scrolling right.
The trouble is that the pages don't look balanced or stylish.
totally agree with Wickham. besides, i still have (so not use) win 98, running on 800x600. believe me there're still a lot of people who use it. and on my own (very slow) pc, i still use 1024x768 (what else can i do, i still have a 15 inch screen =/)
i would make a site optimized for 1024x768, but make sure people who have a smaller screen resolutions (old people use 640x480 :-|) still see the most important things without scrolling to the right.
I think that unfortunately you still have to consider 800x600 but it becoming less and less. I personally just like to make everyone scroll so much until they get sick of it and have to by a new computer or upgrade their old one.

I don't go for 800x600, because why will people upgrade to the better tech. if we don't give them a reason to do so?
Well, most people by now have updated their monitors by now to LCD, unless you are old and cannot see small letters, which most sites allow different size formate for it, or you are in a third world country using the internet, but if they are third world, i do not think they will come to our site

Well you are using a mac then? Aren't you? Mac OS are not really design for full screen applications like Windows or most Linux OS (Since some tries to resemble macs)

Macs can handel full screen apps. i regularly do use full screen apps, such as photoshop, dreamweaver and remote desk control.
BUT macs are designed to do more than one thing at once, which is easier when things arn't full screen. at the present time, i am surfing the net, watching TV, on MSN, on skype amd reading e-mails.
i don't think it is down to a mac or windows or even linux to whether you open things in full screen but down to how big your screen is. obviously the larger the screen the less chance you would need to make something full screen but if you have a 15" or 14" then you would need it full screen.
p.s. i so badly want a mac!

If your screen was quite large it would probably be unnecessary to have an app go full screen. All personal preference tho
